Description of Research
Dr. Goetzl's laboratory is investigating non-migration effects of the lipid mediator sphingosine 1-phosphate (S1P) on T cell cytokine generation, that are transduced by T cell type 1 S1P G protein-coupled receptors (S1P1 Rs). In a TGFb and IL-6 environment, the S1P-S1P1 R axis evokes differentiation of Th17 effector cells from naïve CD4 T cells. In contrast, the S1P-S1P1 R system promotes conversion of naïve CD4 T cells to adaptive T reg cells in the presence of IL-4 without TGFb or IL-6. The T cell signaling pathways involved and the consequences for immunity and autoimmune disease potential are being studied in S1P1 R transgenic and knock-out mice with a range of selective pharmacological tools.
